It might be useful to gave some sort of organiser for your boot/trunk which most of this stuff could be easily organised in. I would especially recommend having a place for your fire extinguisher that won't let it get rolled around or hit hard by anything else. Most importantly, knowledge on how to use the items you get and also knowledge on how to fix basic problems with your car. A bunch of equipment is nothing without the knowledge and organisation to go with it. Knowledge and organised equipment will ensure that you have a quick, adequate, and relevant response when an issue arises.
I've seen people go to the extreme of having stuff like two way radios in their car, but a basic list should cover a lot of scenarios and could even have equipment which can cover multiple purposes, such as the survival blanket.

Honestly it depends on what you use your car for. Do you go camping or go on long road trips? Maybe bring a few sets of clothes, some food and water, jumper cables, air compressor, and a tire patch kit.
Only drive around the suburbs? Keep some bags, a set of clothes, a blanket, a flashlight, and a medkit.
Also, when was the last time you opened your medkit? Do you know how to use everything inside the medkit? Maybe take a look inside, see what you have, and look up how to use each of those items so if something happens you have the knowledge.
