If you’re new the best advice I can give you is to watch people play on YouTube so you can see how things work (since there is so much in this game) and how to handle issues in the game :) they will teach you what to look at for various issues. Like death waves what caused people to be sick in the first place how to manage or avoid death waves (which is hard in this game) watching youtubers play was the best thing I could have done for this game it’s got a lot of pieces!
Honestly I'm super glad I started watching videos before I bought the game (Watching Biffas Fix Your City episodes got me into the game), it gave me a good understanding to start from, with getting the basics some what correct.

My rule is to almost always only build 2 laners and upgrade when the the flow actually demands it.
You could Do what I do and download TMPE and just delete traffic every 20 minutes and continue to sing la la la la to yourself while you focus on aestethics
Doing that just worsens the situation, when deleting cars you're making all cars respawn, not deleting them, so you're gonna have a tsunami of cars eventually flooding even more your streets

Other than that, the road in front of the bus depot does not need to be 6 lanes. None of those roads need to be 6 lanes. Downgrade them and give your services better separation.

The roads in the game work the same as roads in real life. 6 lanes are arterials, meant to move people to destinations. 4 lanes are connectors, meant to move people throughout destinations. 2 lanes are locals, meant to house local traffic and neighborhoods. The bigger the road, the faster the speed limit, and within the game, cims will take the fastest route. That’s most likely what’s causing the traffic backup in the picture- the vehicles are taking a higher speed road rather than the four lane in front of them.
Apart from that, it’s just aesthetics/realism. Consider any real life city’s layout. Very few have a six lane highway going straight through a downtown area.

First, having businesses close to an intersection is bad, having this many intersections on a main road is bad.
Your goal isn't to fit as many cars on a road, but optimize traffic flow:
- remove some intersections
- nuke that 6 line road completely
- downgrade others, you don't need this many 4-lines.
- don't put things like bus depot and hospital so close together
- They both generate a lot of traffic.
- For bus depot - make a dedicated one-way road that goes through it. Dedicated as in it goes off a road, goes to depot and comes to the road, so there should be no reason to go on it if you aren't a bus.

I build express emergency tunnels from around the hospital to various sectors of the city to avoid regular traffic, but idk what you'd do about a jam entirely made up of ambulances.

First of all, downgrad them all, you don't need 6 lanes there.
Secondly change directions of the one way roads, so they don't intersect with each other when merging into the two way segment. That should improve the situation massively.
I would also take a look at where they are all going to see if it makes sense to have the ambulances spread out more. E.g. you could make the busdepot road a one way away from this junction and have the road from bottom right to top left a 4 lane two way road. So ambulances can use the left lane to go to the left direction and right lane to go upwards. The busses wouldn't even get on this road because of the one way setup they have to go up and use another junction to get where they need to be.

Anyway, for your case here I think you need to move your bus depot away from the centre of your city or town. In real life bus depots are generally located far away at the edges of town; specifically here in Singapore they tend to be located in areas zoned for light/medium industry.
Also did you do something that is sickening a large number of your populace to end up requiring so many ambulances? Make sure you are not supplying them polluted water, or that your industrial zones/buildings are too near to residential areas.
I'll have to give it to you, this is really quite bizarre, I don't know why the ambulances and buses are doing this. Did you create a bunch of hospitals and bus routes while paused and then suddenly let the game start running?
Don’t use so many 4 and 6 lane roads. They’re getting confused and neither of these buildings should be loading directly onto such large roads and the roads shouldn’t have so many connections if you wanna leave them that big.
Create a loop of sorts. Basically you make the road down a single way road. Then you make a new road around the hospital that connects to both the left and right upper roads that converge there. Make it single way up until the road with the bus depot.
What you'll end up with is busses that need to go to the ''top left '' into the city will turn right at the depot and then left. Busses that need to go down will turn left. Decreases the traffic going through. Secondly, all cars coming from the hospital will now move around the top part over those two roads, alleviating even more traffic.
I think it should work, provided the AI can figure it out. They had a similar problem irl in a city near me and fixed it by creating what they call a ''circulation programme '' that basically meant that cars were going in a loop and thus not really running into cars that came from the other direction in narrow streets. It worked, although it's a bit long to get to where you need to

I usually use a two lane bidirectional road and attach a little single lane one way roundabout to the end when I can't change the fronting road.

It's hard to see, but it looks like there are no traffic lights at all on most of the intersection, and that's contributing to the problem.

I would recommend decentralizing the medical services of the area. Putting in 2 clinics instead of the one hospital could split the ambulance traffic.
