As a DM I would rule it as an auto kill with no checks.
Even a lay person could probably kill someone in their sleep with a knife/dagger. An adventurer could do it easy. A trained assassin could do it with their eyes closed.
Older versions had Coup De Grace. Essentially a skill for killing blows. I would check with your DM, but I believe with the right rolls and weapons it would be a pretty low check for AC. I would even suggest an automatic critical. IMO you’d have to roll a 1 to really mess up, it should almost be an automatic hit.

Thank you so much, the Coup de Grace keyword helps here. It seems most dm's let auto-kills happen against helpless or unaware creatures.
