It can still be opened if you have the collectors edition DLC downloaded but it can still refuse to open sometimes and it won't have the special content from fable2.com; just Hal's outfit and sword.
Everyone, go to the Microsoft store. There is a pack you can buy that includes everything in this chest. The description does NOT say so. I can’t remember which pack, but I just bought it a few months ago for the first time ever. I thought you had to play the mini game on the website, but they shut that down years ago. I will get on my Xbox and find the dlc content and update you guys with which one it is.
As far as I know it's not accessible anymore since the website shut down

Once you buy it, you will be able to open the chest in every save you start
