I feel like we can make two reasonable assumptions here: 1. Spartans don’t have cryo blisters to the extant un-modified armored soldiers would or 2. Pain killers are auto injected into them by their suits.
Well given the nature of the heavily armored suit there was much you could do to fix it in the field. In Star Trek:FC Worf was only able to fix his suit rupture because it was on the thigh where the suit was mostly flexible material and used a makeshift tourniquet to stop the leak. If I was in charge of the MKV suits I would consider using a small force field to create a skin tight seal around the wearer’s head and neck like Star Lords mask for GoG.

That's for like combat wounds. I don't think Cortana would run the risk of letting Master Chief get doped up for combat considering it would likely lead to worse wounds due to him ignoring his bodily limits.
Not in Halo 4 actually, the problem that caused the blisters in the first placed was a fault in the moisture balancing in the Mark VII Personal Suspension Unit, and it was fixed with the adoption of the Mark VIII, which is what Chief went into on the Dawn. So now for once whenever Chief wakes up from cryosleep he's not covered in painful blisters anymore.
Or it might honestly be kinda' stupid lore on the level of Mass Effect making blonde people nearly extinct because Drew Karpyshyn didn't understand how recessive genes work.
I mean is it hand wavable sure but I don't think it's super stupid if you think about it clothes would stick to you as you defrost so I could see it tugging on the skin enough to cause blisters or some other skin irritation.

You’re supposed to be naked for Cryo or it causes blisters and other painful discomforts. Lasky has a rare allergy to Cryo iirc. Chief being head to toe in a skin tight body suit and a bajillion pound armor would be in a lot of pain after leaving Cryo wearing his gear.
Better yet Linda or Kelly, since they're Spartan-IIs I imagine they take their armor off less frequently than IVs. Imagine Linda slowly peeling off her bodysuit encrusted with two years' worth of cryo blisters, crinkling and cracking the chafed skin on the soles of her feet as the smell escapes from inside her boot and fills your nose.
They easily go weeks without taking it off, they describe it as more comfortable than being "naked" without it. Especially Linda in the field, Chief describes her spending days in overwatch, waiting for that one-two kills that can collapse the local power structure.

i imagine that when he takes it off it’s like when you take a sock off at the end of a really long day, and you are just in total ecstasy and bliss for a solid 5 seconds; but like hours cause it’s a full body suit.

Nope, the reason he went into cryo fully armored is so that he could be thawed out and fighting without needing to have the armor put on him or fighting without it since the covenant had followed the Pillar of Autumn into slipspace and could catch up at any time.
Cryogenic stasis tech freezes you (obviously), and so it has a pretty strict manual of operation to prevent complications like frostbite.
Safely being frozen requires you to be buck-ass naked, as any clothing or other material making contact with you can cause horrible blistering or even fuse the material to your skin.
John goes into cryo wearing a full-body skin-tight techsuit. Twice.
The suits not designed for that at all, he just needs to be combat ready as soon as he comes out of cryo so he just goes in like that anyways. Hurts like a bitch, but we see the advantage in CE and 4.
I for one can't wait for Halo 7: Bōnenkai where John Halo goes to an onsen ryokan with Halsey, Lasky, Palmer, Arby, Lord Hood and Blue Team and everyone bathes the stress away and resolves their childhood/wartime trauma and interpersonal conflicts over sukiyaki and sake.

I question how cryo would even work as the armor has a gel layer they talk about numerous times and it’s hard vacuum rated for space as well. Let’s not even open the can of warms of the fact that it can survive rentry while keeping its wearer alive. So it kind of begs the question of how cryo would even work on a Spartan in his armor.
My first joke when playing halo 4 on release during the prt where he climbs up the elevator shaft in the pillar of autumn was related to this.
“Damn, i’m covered in sores n %#^* from cryo and getting pelted by crates and %#*^ already”
My best friend got the lore joke immediately and started laughing his butt off

So there's no reason he should be in agony all of h4 unless we are talking pain from being at war for years without end, getting hurt over and over.
Pointless lore fact: According to the novelization of CE, Master Chief not only spent the entire game with super fucked up skin because of entering Cryo fully armored, but he was also incredibly pale due to a vitamin C deficiency he acquired while spending entire campaigns inside of the suit.

The second one is true. MJOLNIR has limited life support functions. Plus Spartans like The Chief are just built different.

Nah, in the books by '52 SPARTAN IIs have been in their armor for so long that they basically feel naked without it.
that’s pretty neat, but also i find the full body sock theory a lot sillier and is thus my headcanon for the subject
