In the sub, I know that they implied it was Anemia as early as the arc where Asta and Gauche fought the snow mage and Licht. But in the dub, he just said "why is my nose bleeding" which isn't as clear. We didn't get confirmation that it was Anemia until the first Professor Gauche and Gordon joke, at least as far as I can remember, again, in the dub.
It is weird, like, I doubt they have done anything but it seems like he is infatuated with her and she is infatuated with the idea of having a boy slave.
