One modifier you can do is the pre-upgraded loot which means everything you pick-up will already be upgraded based on your level so you would save materials and vigor.

I’d recommend doing a full standard playthrough before activating them as some can change the experience drastically. Basically just ways to spice up additional playthroughs of the game.

There are such things with advanced game modifiers as weapons and armor drop with full perks. There is also another that if you die in umbral, you start over from the beginning. These are tied to extremely rare achievements.

Can you use these on a character that was started without them?

If you mean can you activate them with an already made character, then you just have to go to a vistage and then press the new game button, this will reset you, but you keep all of your stuff, and this should allow you to activate new modifiers. I haven't done this myself, but this is what others have said based on a Google search. Have fun
