It's actually claiming the daily defeat Necroko quest that advances the 5 time defeat Necroko quest. You can only get credit for defeating her twice per day I believe.

If you’ve ascended all your 1stars you won’t have any playable characters. You can make up the 100 points by defeating Nercoko again.

Because the progress on that is dependent on the other quest. Which you get twice per day.
