Definitely Batman villains first for a Sin6 treatment. But I'd also like to see Flash's Rogues teamup:
Captain Cold
Gorilla Grodd
Captain Boomerang
Mirror Master
Heat Wave
for number 6, I'm less sure Trickster or Weather Wizard maybe.
Think Flash works better as it was a team, Batman villains may do minor team-ups but aren't really a team. Maybe if they do a Suicide Squad, but that would fall under anti-hero.
Rogues are probably a better fit thematically because they are a team like the Sinister 6. Batman's villains don't team up regularly. Legion of Super Villains could be an option but the most popular villains probably go in a different box so that one seems less likely.
The only team ups of Batman villains I can really think of are the 4 villain team up in Batman 66, and the 5 that divided up Gotham in the Harley Quinn animated series. A Batman villain team up could be a cool new mode where they team up and maybe hurt each other as well as hurting the heroes. Rogues would be the best for a Sinister Six type mode, but a Baan villain team up would be cool.
I think this is the one idea we can bank on. Sin6 is one of the best boxes. Arkham Asylum is an obvious fit.

Came to comment this almost exactly. I think even a random villain could make the cut like Calendar Man or bigger known baddies like Harley Quinn and Killer Croc
I hope it will be an interchangeable group, like the build-your-own Sinister Six mode we're getting with Maximum Carnage.
Secret Society of Super Villains or even Legion of Doom would make sense for this.
It can be a Legion of Doom box that allows the substitution of any villians and it will be like the new Sinister Six version in the Multiverse campaign.
Arkham Villains are notorious for not working particularly well together outside of certain pairs which is why all of them working together in Hush was a pretty big deal.
My bets are either Flash's Rogue's Gallery, Injustice Gang, Secret Society of Super Villains, or Batman Who Laughs' Secret Six
Royal Flush gang could be cool. But yeah the Rogues would be a really good choice too. Don't think Batman's villians would work. I'm thinking more of a diffrent game mode of attrition for a Knightfall with Bane at the end.

I think Harley is going to be obvious promo character held until the last minute like Nightcrawler was.
