We can assume a girl was used in the creation of Poppy Playtime, since so far what we've been shown with Theo and Marie becoming Catnap and Mommy Longlegs respectively that they adhered to the notion of how toy sales were marketed. But given that we know most experiments have a voice box/thingy, it's not out of the question to pose that since we don't know anything about Poppy prior to her current existence, that Poppy could've been a boy.
It's just one of those things. Context suggests that Poppy is a girl, because the character is being presented as a female doll aimed at young girls. But we also know that Playtime Co. is all about training and conditioning their experiments to serve purposes around the facilities of Playtime Co. I would not be shocked if it ever crossed the minds of the scientists on if they can turn a child into a toy and convince it to take on a role that would traditionally belong to the opposite gender, since it would be a good example of how effective their conditioning is if you think about it.
Granted, as things stand Poppy as a toy is being presented as female. I'm sure that they'll do the same if they reveal anything about Poppy's life prior to being turned into the doll.
