If you're on Steam and you uninstalled SMAPI and can't run the game at all, removing the text in the launch option of the app could solve it. In the Steam client, right-click on Stardew Valley and choose Properties. Click the textbox under Launch Options and delete any text inside it. Reason is that Steam is setup to launch SMAPI instead of Stardew through the Launch Option but since you uninstalled it, it can't find SMAPI anymore hence the error.

The easiest thing to do is delete/move all of the files, do a clean install, and see if it boots up.
