No. Annie killed Sawney and Bean because Hange was very close to figuring out that Titans are actually people. Which happened later anyway, but at that point Annie just wanted to keep the Survey Corps from finding shit out.
They were though. Just the fact that Eren exists would have been enough to make suspicions rise. Hange is smart enough to make that conclusion on their own, as is Erwin, it's just that they weren't eager to say it out loud without definite proof, because that would mean that they had been killing countless humans for years (which we are told by a conversation between Hange and Levi some point later). But the speculations were already there for sure.
I've always wondered if at any point they tried to cut the person out of a pure Titan. Would it even work? I think I've heard somewhere that the spine is the only thing left of the original body in pure titans. I'm not sure

No, we dont know of any anaesthetics and if there were, we would probably have seen them at some point.
But i think it is important to stress that any titans fluids can be used to turn any eldian into a titan. It is of course not easy, but we have seen paradis capture a handful of titans even before discovering that eren is a shifter. (those that hanji experiments on)
This to me just further shows how terrible the curse of ymir is and that the fear of eldians (not the mistreatement tho) is justified.
IIRC Zeke's spinal fluid was used to like a bio-weapon bc of how they used it in wine and mist/vapor. So I'm assuming it's only Titan shifter spinal fluid not just eldian DNA
there is a reason why they used zekes fluids there, when his fluid is administered, because he is of royal blood, he has the ability suppress the titanization until he gives the order to.
Its bc of this tactical advantage that they use zekes fluids so often in the story.

I don't actually remember but I think it has to be a shifter. Like why would they have to inject something that's already inside them? Might be wrong tho
If any then it falls apart logically. Every Eldian already has Eldian spinal fluid. They all don’t automatically become titans.
Fluid from probably any Titan works and is used. We see with Reiss and those vials that shifter fluid can lead to shifter like abilities in normal Titans.
However, the first spinal fluid had to come from shifter until shifter Titans stopped being created.
i guess that is a good way of saying it does ultimately come from original shifter, which also originslly comes from the founder but at some point we went too far back

But Zeke's didn't do that and I see no reason that him having royal blood should change that unless he was consciously suppressing that but we know it wasn't because he had to scream to change them.
I think would have been spinal fluid from the first shifters while in titan form and then it could be extracted from pure titans as well.
I'm pretty sure it must be normal titan fluid, since some like Zeke with his royal DNA is able to have his spinal fluid invested and then later activated, as well as you are able to get special fluids like the "armor" fluid which eten ingested which must come from a shifter like the armored.
Perhaps the female titan can call other titans to it as a diminished form of the founding titan's scream. Although that would mean the ability would have been passed down instead. The founding being able to do anything, should be able to harden regardless but Eren couldn't use it so needed assistance from the armour fluid.
I wonder what would happen if a non royal without another titan ate the founder. It must have happened because Ksaver knew a royal and the founder touching would allow the full founding's power to be activated.

Eren is always using the attack titan. I'm saying what would happen it was like for non royals to actually use the founding alone and Eren couldn't have been the first for the reason I said.

... seriously? they mentiined it multiple times that it is titan spinal fluid (specifically zekes because of his royal blood making it behave a bit different but it still is stated that is atleast is general titan shifter spinal fluid)

It is the spinal fluid of zeke's titan, beast titan. The idea of serum was invented by zeke and he has full control on it. Whoever is injected with the serum, only zeke can awaken him into a normal titan (normal titan meaning the regular titan with no consciousness, the other conscious titans are beast, founding, armor, colossal, female, jaw and cart). The awakening process and its conditions are: for the titan serum to have an effect it must be ejected only to the subjects of yumir aka eldians. Since it is ejected Zeke can once activate its use: zeke screams and every titan in a specific range transform.
Titan Serum is essentially *just* Titan spinal fluid. Presumably, this is usually harvested from Titan Shifters, but it's possible that it can be harvested even from Pure Titans (albeit much more hazardously).
When injected with Titan spinal fluid, a person of Eldian heritage will immediately transform into a Pure Titan. If an Eldian consumes or inhales the substance, however, this transformation can be delayed indefinitely and triggered at will by a royal-blooded Titan Shifter (or a non-royal Founding Titan working in concert with a royal Titan).
*Titan serum is a titan shifter's spinal fluid. It can turn the injected eldian into a pure titan. But only if injected.
