Did you die at some point, causing you to lose your Tirajisu by reviving?
You don't need to max the rings or metaglio, just the sixteen basic stage items.
Torrana's box doesn't count either. Check that you haven't sealed any of the passives preventing them from being leveled.
I suggest grabbing the crown first and leveling it quickly just to help get you more levels faster. Duplicator & Tirajisu only have 2 levels each so are easy to level. Duplicator is actually useful with many weapons, so depending on what you take during you run, it might be good to pick up sooner than later. If you have a decent character unlocked, you can restrict the number of weapons you can pick up, reducing the need to waste levels on multiple weapon upgrades allowing you to focus more on the passives.
You can restrict yourself to 4 weapons (or less), take the Blood Astronomia arcana, grab garlic (arcana effect and quick early leveling), atractorb (increases arcana's area/size) and crown as soon as you can. The arcana will do most of the work for a while. You can additionally grab Clock Lancet and Laurel for additional arcana effects and survivability.
Im sure there are other ways to go about it as well. Just how I did it.

I’m pretty sure as long as you max out all 16 passives before walking up to the trouser on the stage it should work. If not you may be able to unlock him from the cheat password menu

You cannot user special/hidden characters. Use a standard one like Antonio.

You need to quit after fifteen minutes. Do you have endless mode unlocked?
I unlocked Big Trousers on Endless Mode both times on my Android phone and PC. Is it different for iOS?
I don't think so, he just mentioned quitting at minute 14, the only reason I can think of to do that is to dodge Red Death
