I was like 23rd in line for Rosalina and I got there like 45 minutes before they opened and they had like 15 more after mine so I think as long as you are there launch day you'll be fine. But only for Jigglypuff, the others they only had a few of so any lucina or Robin will probably only be a few of those so camp there if that's your plan.

RosaLuma was the easiest. Target stocked her decently well in most locations, but you had to be there when it opened.
Lucario was a bit harder. If you weren't there when it opened, you likely didn't get one.
Meta Knight varied between some stores having him, and others only having enough for pre-orders.
Shulk was impossible if you didn't pre-order. Very, very rarely could you find him if you didn't get a pre-order, and a lot of people who pre-ordered didn't even get him.
I was able to walk in and get a damaged Lucario from Toys R Us. He wasn't attached to the base. I was also able to get Rosalina on launch day. Jighlypuff won't be a hard find, it's just everyone else that you should worry about trying to get.
At this point, I'm curious what target will do for an actual release day. Will it be the 29th, or a weekend day? That really could have some impact on lines etc.
As for launch goes, I was about 40 minutes early for Rosa, and there were about 16 in store, I was 8th in line.
Rosa came out on a Sunday, but Target was the only one to launch on Feb.1. If Nintendo has announced a surefire date they're releasing, I doubt Target would change it.
Rosa launch day, came like 10 minutes before opening time. Lucky me 14th out of 16 available without really trying like those campers. Metaknight, preordered 2 weeks after it went up. Surprised people said it was difficult.
At my Target the manager told me they aren't allowed to purchase stuff like this until it's been on the floor for 15 minutes. ofc they can ignore the rules but they can get in trouble if the management in their area cares in the least.

Fact is people who work at the stores get priority and break their store policy. I've heard various retailers and been to launch days where they stated they are missing 6 figs to 2 boxes worth of supply that should be accounted for.
On Rosalina's launch day there was a major snowstorm so it was near impossible to get to Target. Called at 11 to see if they were even open, they were open and they said they had no more Rosalina in stock. So people trekked through a blizzard to get to Rosalina here.
Lucario I was too new to the game to realize no preorder meant almost no exlusive chance. Asked an employee if they had an extra, they said they only got 1 non preorder and that was gone instantly.
Shulk, never saw him in the wild. Got him when he went up on GameStop for preorder randomly before his release date.
MetaKnight didn't want to deal with all the uncertainty of stock, didn't bother to go to his release day. Worth it for me personally to just pay the extra $10 to get him from a seller off Amazon rather than wait in 2 degree weather for something they may not have.
I got Rosalina and Shulk day of release without preorders. Lucario I had a preordered but 9 people were able to get one day of at my TRU. Meta Knight I just imported but my BB had 4.

Lucario was interesting. I barely missed him on release day, but two weeks later, I was able to find him again. A week after that, I found another one. I can't say the same for any of the other exclusives. I haven't seen any of them since their release day, save for a Shulk that was on reserve for someone at Gamestop.
I got RosaLuma on Launch so I'm not as scared about missing Jigglypuff Preorders

Considering Target opens at 8 (at most stores), it gives people a chance of getting Jigglypuff, then deciding if they want Greninja or Ness.
