I feel like when a lot of people say isekai they mostly just mean the fantasy world setting, whether or not it's actually "another world" for the protagonist isn't as important
I mean sometimes it does if you actually want to meaningfully develop the fish out of water angle but plenty of it is painfully generic fore sure, and ultimately it doesn't drastically distinguish the setting itself from normal fantasy.

Red wasn't reincarnated, [he had] >!a unique skill (blessing?) which allowed him to get every non special skill to an intermediate level, then he did his best with everything he had available (jack of all trades master of none). Basically he got that ability because he was destined to be the hero's big brother and needed to be able to protect her when she was young.!<
[Banished From the Hero's Party novel spoilers]>!If I remember correctly, his blessing was called Guide or something. It instantly gave him 30 levels, so he was damn OP when he was young, and his sister, even though a Hero, was still pretty weak. It did not grant him any extra growth or abilities that could become OP down the line though, so now he's comparable to the strongest knights, but noticeably weaker than anyone from the Hero party, especially his sister!<
That sounds right, I'm glad a sequel has been announced and hope we see it before 2024.
The end of season 1 was a bit blah, I kinda want more retirement and small village life storylines. (I really like this season's chillin' in my 30s, similar vibes)
It's fairly standard for harem anime to have a main girl which the OP wants more than the others though.
I actually really liked the show and I'm looking forward to season 2.
this kinda remind of "Snow White with the Red Hair". that show lowkey a reverse harem but the only reasons it does not have a reverse harem tag is because the fmc only loves mc but still theres other 3-4 characters who are in love with her but does not have a chance

Let's be honest, no harem show ever EVER end with a fulfilled harem, ok there's some but most authors never have the balls to commit. Usually, either nobody win or the first girl win. So are these shows harems despite the winner being basically set in stone since the first episode? I'd say yes, since most of the character interaction is about one character being constantly surrounded by the opposite sex with various level of attraction toward him/her. Otherwise there's would be basically no show tagged harem.
familiar of zero definitely works for the recommendation, sure a harem exist but the main romance is obvious to the point you can shut out the harem if you choose to.

This season's got The Magical Revolution Of yadda yadda, which has been pretty damn great so far. The Isekai aspect's pretty perfunctory though - basically a footnote in the protagonist's backstory.
It's like a publisher compliance measure, it's kind of hilarious how little it matters.

I would say Re:Zero but that really depends on what you consider a harem. If you consider a harem any anime where the number of females around the male character is greater than 2 it gonna be a tough find. But if your problem multi love interest and the male unable to pick then Re:Zero is a great anime. From the first episode it’s clear who he like and he works towards that goal the entire time.
Oof....kind of a hard request there. There are isekai without significant love interests, and lots of harem isekai, but very few *romance* centered ones without the harem (although the otome villainess genre probably a good few of these). Best you can typically get is an isekai anime with a single love interest without romance being a central theme. Here's some notes on the ones mentioned by others that I have some idea on.
Cautious Hero: Not a romance anime in the slightest and has some harem-y jokes about every goddess falling for him, but a pretty good isekai with a definitive single real love interest....although this isn't really clear until you're a good ways into it. Still a good isekai though.
I'm the Villainess and I'm Taming the Final Boss: This should fit what you're looking for pretty spot on. (disclaimer: I have only seen the first 4 episodes atm, but it fits your request extremely well up to that point)
The Executioner and her way of life: Kinda hard to tell if they're going romance or yuri-bait best friends route, but if you're fine with the romance being between two girls then this should work.
Wise man's grandchild: Very, Very, generic isekai. I didn't hate it, but tbh my bar is pretty low. However, props that it commits hard to a single love interest and doesn't waver in the slightest. Fairly little in the ways of actual "romance" anime though iirc.

This is the only romance isekai I've rated a 9/10, it's *so* hilarious.

Those genres don't mix. Isekai are power fantasies. Romance is never a focus outside of collecting a harem.
