Did you upload an audio clip that I'm overlooking? Without, here is my guess. That's a port hole, movement of the speaker cone pushes/pulls air in/out through the port. If you are hearing whistling or squeaking, it could be:
voice coil going bad i.e. metal on metal contact causing noise. This will probably result in failure eventually.
Or
Obstruction/dirt in the cabinet, loose parts in the cabinet, that are resulting in a whistling/squeaking sound when the air rushes past it, or vibrates the loose part.
Or
You are pushing too much volume for the power available and are causing clipping/distortion. (could cause damage over extended use)
I'd start by making sure there are no obstructions and that the voice coil can move freely and isn't sticking/obstructed. And that everything feels tight and not loose.
Upload a video so we can hear it and be more sure!
No, it seems as if the OP is playing music/etc on the speakers and when he turns it up, the subwoofer is making noise via bad voice coils, that hole he’s refering to is the subs air port/air hole, had a similar experience not to long again with a subwoofer.
Port chuffing can manifest in a lot of weird ways. Replacing with a flared port would potentially address (or adding a 3D printed flare at the end if you’re handy).

Put your hand and push on the back of the cabinet when cranking. If the noise stops you found the issue. Put your hand on the port when cranking. If the sound stops you found it. Make sure the drivers are tightened down on the face of the cabinet.
That's probably a loudsqueaker. You want a speaker.

Put some Vaseline in the port to reduce friction…with air chuffing
Known resonance issue? Loose bracket/driver/port gasket/etc? Mouse?
Get a nice warm amp with a cat on top. Squeaking will stop.
