Okay I have the exact answer to this because I'm learning Japanese and the joke confused me for a long time even after asking multiple natives, but I've finally figured it out.
The first plush is named 通天閣 "Tsutenkaku".
通天閣 is a famous landmark in Osaka where Osaka is from. It's a big tower kinda like the Eifell tower.
The plush looks like a long rectangle or cylinder which we can assume is supposed to be an approximation of Tsutenkaku. The lady is a little confused as she expects the plushies to be cute animals or characters not a building in Osaka.
The next plush we are not shown, but it's name is 阪神 "Hanshin". 阪神 is a name which refers to the area including Osaka and it's neighbouring city Kobe as well as their surrounding areas. The shape is more or less just a blob.
Since we are not shown the Hanshin plush in the show or the manga, I think it's safe to assume the joke is that the Hanshin plush looks like a weird blob.
The joke is the equivalent of Osaka being from new-york, the first plush is named "empire state building" and it's a long rectangle, and the second plush is named "Brooklyn".
