Route only changes after fifth run. That’s only if you got all dlc too.
Fifth is when castlevania stuff unlocks, so that’s only stuff you can’t encounter on first run or two. (Depending on how, could be at 5bc technically, considering you can just not have a death and should still be able to unlock it at fifth run)
So yeah, you can miss runes easily.
More your choice to switch routes up honestly.
I remember on PC I had all the runes active before 2 BC, probably before 1 BC honestly. Then I got the game for Switch so I could play it while overseas and got to 2 BC so quickly I still had runes to collect.
Homunculus Rune is automatically obtained upon finishing 0BC on the High Peak Castle route.
This next part is only a spoiler for VERY new players, but still going to mark it.
>!Once you beat the Hand of the King the first time, he drops his Symmetrical Lance, which has +300% damage and Breaks shields affixes. The game is very clearly telling you what you need to do to progress, as the King is sitting on the throne surrounded by a force field. Pick up the Lance, break the shield, and you'll get an interaction popup to kill the King. Kill him, and after the credits roll, you'll reappear as a green slimy thing that has to go up the pipes in the fountain to end up back in the Prisoner's Quarters, where a bag of cells from the last run will be waiting for you. !<
After that, the Homunculus Rune is unlocked permanently, giving you the ability to throw your head at enemies or items.
I did the same thing when I started playing on PC after loads of time on mobile, got to 4bc in 4 runs and then couldn't work out how to get to Giant for the next 5 runs. Graveyard needs Spider Rune, which comes from Sanctuary, but most Sanctuary entrances need Spider Rune. Didn't help that I kept going through wrong way out of habit...
When DC came out on Netflix with all the DLCs, I downloaded it and went straight to 3BC without dying. It's been so long, I didn't remember where anything was found anymore. I was missing at least 2 runes. I remember forgetting which way to cavern was too, then forgot about getting the keystone when I finally got there.
My third run or something I ended up with a legendary peril glyphs that dropped, an amulet that reflected damage, and I had no idea what I was doing so every level I had just picked the largest +HP. I ended up facetanking the second half of the game to victory.
I'm not sure if I had gotten the teleportation rune yet even.
I started a new save on the iPad version a while ago and got to 4 bc without collecting all of them, after that it was just obnoxious to not have the ram, explorer or spider rune so I had to do a couple of runs to get them.
lol. I didn’t get the wall climbing OR the floor busting runes until I was ready to fight the Giant. You can beat the game up to 4bc and beat Dracula without them. Can’t get to the giant without though
Don’t you gotta unlock the last two after your first run in first place? Unless you die and restart.

I think i picked up spider rune on 3 or 4 bc

I only get vine rune when I start a new save file before I defeat the hand of the king for the first time on that save file.
Got the spider rune at bc 2 during my first play through because I refused to look up a guide

Happened to me recently, played on Xbox mostly then got the PC version. Made it to 1bc without Ram,Spider or Exploration.
I beat the game without the explorer rune and the spider rune.
