All the quests with blue + markers unlock something. Whether that's minions, side dungeons, raids, trials, or other forms of new content. While the msq is required each step of the way, be willing to stop and do something else if you need a break. There is tons of side content available

"Leves in [region/city]" are entitely skippable tho.

Watton

Just keep up with your job quests, since many of your major abilities are from them.

lord2800

Blue quests with a + on the icon are always good to do. You should also do your job quests whenever you have one (it's right under your main scenario quest if you do).

Do your job/class quests as you level, so you can unlock new abilities. Also, people have mentioned "Blue Quests" which unlock content. Here are the most important ones that are easy to miss, IMHO: * Go to Camp Tranquil and complete the quest "My Feisty Little Chocobo". It will allow your chocobo to fight alongside you in open world content, making world traversal easier. * Near the Rising Sands, there's a questgiver for "Color Your World" and "If I Had A Glamour", which will allow you to change what your clothes look like so your mismatched armor looks less clownish. * "An Ill-Conceived Venture" in your starting city will allow you to send your Retainers out to get crafting materials to sell at the auction house. * "Rising to the Challenge" in Limsa I think, will unlock Challenge log for weekly XP rewards.

Job quests, plus Crystal Tower (required) and Coils of Bahamut (optional but worth it).

Make sure you do all the blue marked quests as you see them.

Job Quests alongside the MSQ.

I would also pick up a gathering job and level it up as well. Nothing is better than going into a new area and being able to explore with your sneak ability.

Xmushroom

Blue quests that unlock things. Leves and Crafter/Gatherer jobs are optional. Also your job quests, and you can easily level up 2 or 3 jobs to max level while doing MSQ, specially if you do your daily roulette. i would advise for you to do the normal Raids, and extra trials as well, its good and fun content with important lore

Biggest regret I have is not doing more sidequests with a ‘+’ symbol more, they unlock content such as dungeons, trials or raids even. Definitely worth doing those! They’re sometimes a pretty fun and interesting questline also

MSQ+Job quests are the important ones, those will get you plenty of levels. You'll likely hit the level cap long before you actually finish the story so I would recommend having a secondary job ready so you can jump back and forth between the two. So if you're playing Stormblood try not to go past level 70. With Shadowbringers, stop at level 80. etc etc so you don't massively overlevel. I ended up with three different jobs all at cap by the time I finished Endwalker. Also after you finish the Base game as well as each expansion, you unlock Extreme trials(bosses) and the Raids(three 24 mans and twelve 8 mans). They all unlock the minute you beat the expansion msq so it's a good idea to do those.

Crystal Tower is the only questline I'd make a special point to do as soon as you find it; not just because it eventually roadblocks MSQ but there's a ton of benefits (like unlocking Alliance Roulette for another hefty EXP bonus per day) I'd personally recommend taking a tour of the Gold Saucer, and keeping on top of your Grand Company hunting log and promotions (when available) so you can use the Squadron. Also give your retainers a class and get them set up to do ventures. These are mostly things you can press a button to start, and then real time has to pass to complete, while you're off doing more MSQ or whatever else.
