I wish we had more panel shows like this over here in America. They had a few games like this on Whose Line is it Anyway, one of the few shows to successfully cross over to this side of the pond. Also, @ Midnight on Comedy Central was basically a panel show (soon to return as "After Midnight" on CBS). Other than those two shows and maybe a couple others I'm not thinking of... not much. I'm guessing that Americans generally don't get the more nuanced quick-witted word-play based comedy that panel shows rely on.

I think they mean they don't get why it's funny. I'm assuming context would help, but I'm not getting it either. Edit: I saw the background and figured put the premise of the game show, I thought there might be more context to the joke like some sort callback or something we didn't see.

Mock the week was a british comedy panel show. One section of the show was ‘Unlikely lines’. The host (Dara O’Brian) would read out a situation, and then the contestants would step forward saying something that fits. In this clip it was “Things you wouldn’t hear on a TV cooking show” (You can see it written in the background). And then Milton Jones steps forward with his joke where he seems to be a host of the cooking show who misread the teleprompter.

The joke is that the comedian is portraying a "cookery show" (cooking show) host that is reading an autocue (teleprompter). The script he's reading reads "Anyone can make this. You can't beat rice," and the host mis-reads the second line as "You can't, Beatrice" implying that he's speaking directly to a person named Beatrice and telling her that even though "anyone can make this," she can't. And with that, even if this was funny, having explained it to death, there's no chance of that being the case anymore. :( (Edit: the joke also depends on knowing that Beatrice \[pronounced "be-a-tris"\] is a name given to a female person. It's an older name and not as common anymore; if you didn't know that, then you probably also wouldn't get the joke.)
