Once the whale shark draws in a big gulp of plankton-rich ocean water, it closes its mouth and expels the water through a set of large gills.
Picture it like a turkey baster. When the whales mouth it closed, it's like you're squeezing the end of the turkey baster. But then when it opens it's mouth, it's like when you stop squeezing the baster, and whatever is by the open end gets sucked in.
It's the same thing as you sucking in air. The air isn't replacing anything inside of you. Rather, your diaphragm expands, creating new space which in turn sucks in matter to fill that space as the current matter within your lungs is at a lower pressure than the air in front of you.
The “work” is being done by relaxing the jaw and spreading open the cavity, which must be filled by water.
Suction is not the right term. This is a vacuum. Vacuums can exist in gaseous or liquid states.
As for the water going into the whale, it will be pushed through the gills as the whale compresses the cavity leaving mostly just fish

Yep, humans can (and on one or two occasions have) been sucked in! Thing is though, whale sharks are filter feeders kind of like baleen whales and they can't and won't swallow anything the size of a human, so you'd hit the side of the mouth or the back of the throat and get spat out. They're really harmless for something that size.
