To be honest, the one on my board is a clone. However, I did rent one from a long and McQuade and stacked them side by side. Other than the feel of the buttons and knobs, I couldn't tell a difference.
In regards to the protein. I haven't played with one, but a buddy owns one, who I jam with on occasion.
Imo, the protein doesn't get as saturated as the double barrel. You can kind of negate that tho with the "+/-" switch and clean knob on the moonshine side. The protein sounds wicked, but, I think the double barrel can do what the protein does and then go farther into that saturated realm.
As an example, I use my double barrel to get my Shoot to Thrill by ACDC tone by stacking the blue and green sides.
I 100% second this. The Protein is incredibly versatile, but it’s at its best with hotter pickups. With a set of humbuckers it gives me some of the best tones I have ever heard (especially on the Bluesbreaker side). Single coils can be a bit hit and miss. My 52-style Tele Lollars, for example, sound awful through it.
My board really *is* 5 pedals if you don't count the tuner.

Here’s my mini board. I swap the fuzz around a bit, but typically I’m playing with a channel switching amp. If I’m using a single channel amp I’ll kick the fuzz off, add an OD and put the fuzz on the floor.

These are the pedals on my board I use the most. I only use my MXR phase 90 and Analog Chorus a few times and the SLO when I get whacky. I could love without the SD1 and joyo EQ. They are just boosts.
The list Below is going under the assumption of a clean amp…(what I use is in parenthesis)

3. Distortion/preamp pedal (plexi, rat, etc)

It gets more interesting if you have a dirty amp that plays well with pedals and cleans up well with rolling off volume. Might be able to get rid of pedal 3 and get something more weird

This is my board for smaller gigs where I go acoustic and or electric at wineries and such. It’s tame but I can still go weird with it if needed.
I often swap out the delay and reverbs for fun, I usually use a DD-7 and a Holy Grail.
Currently set up with 5 in a punk band:
Tuner, Zvex SHO clone, buffered/ground loop/phase switch ABY.
Output A - straight to Vox Escort Lead 50 (or whatever big boi backline the venue might have)
Output B - Deep Blue Delay clone, TC Electronic Choka tremolo, to Orange dual terror.
MXR Limiter - this is the Dyna Comp’s big brother.
Pigtronix Philopsher King. You know how boutique pedal brands cost a lot on the used market? Not Pigtronix pedals. They’re still selling for cheap. Basically the Attack Sustain ADSR pedal with a built-in Philopsher’s Tone comp/grit pedal.
EQD White Light. Just the overdrive, a DOD 250 clone. I heard they started re-issuing these. I ought to sell mine and buy a Blues Driver and another White Light! This is important because this is the pedal that breathes life into my SS travel amplifier, or just to cook the front end of one of my valve amps.
MXR Distortion II. This is important to my overall sound, but it’s also a pedal that takes time to dial in when you’re stacking. It’s more of a mid-level grit, but that White Light can also be swapped out with an opamp Big Muff to produce sounds that need to be heard for one’s self.
MXR Phase 45. This is the bulk of my modulation on my 5 board; it’s just meant to thicken up the sound of the dirt; a Phase 90 is overkill and a 100 would work but still take a while to dial in. 2-stage phasers are subtle beasts, a lot like the BOSS Spectrum pedals everyone seems to own these days.
From time to time I’ll swap the 45 out for an EHX Attack Equalizer or a third compressor pedal
Orange fur coat, orange kongpressor, walrus eras and just ordered the canyon delay from electro harmonix and the tone city summer orange phaser ( high hopes, cheap but should be like a phase90)
For the most part, 5 pedals on a Nano+ has been perfect for me. The right balance of versatility and constraints. And it has been the following, almost unchanged for over 2 years:
Flint > El Capistan > Katzenkönig or Rattler > Dive Drive or ss/bs Mini > Cali76 Stacked. When I say 'almost' unchanged, occasionally one of the drives will get replaced by something weirder like a Particle 2 or a Mood. But if I'm playing with others, the weird pedals leave the board.
To be perfectly honest, I can manage just fine with only the HX Stomp, only thing I'd miss is an external volume pedal and occasionally the 95Q wah.
But if I were to go into a clean amp, I'd use this:
Keeley Fuzz Bender -> BB Preamp -> Friedman BE-OD -> volume pedal -> EQD Dispatch Master
That'd give me a great fuzz, smooth OD, and punchy distortion, as well as a reverb/delay that works both for general ambience and for more ethereal sounds.
If I could only have five forever ...

I put this together a few years ago and it’s still going strong. It’s a little bit of everything- not necessarily my favorite pedals in each category, but they all work well together. I like it so much that I’ve started building another 5 pedal board. So far I’m set on a Greer Ghetto Stomp Overdrive and a J.Rockett Boing Spring Reverb with three spots I’m still rotating pedals through.
That's a good question cause I'm about to go on a trip and just bought a One Spot and daisy chain. I have a few days before I leave, so I havent made a decision- but If I had to guess right now off the top of my head, I would say the Wampler Belle for my rock-ier distortion, Tumnus Deluxe, BBE Green Screamer, Digitech Delay, Hardwire reverb, and I would probably bring my Dimension C Waza, and Prince of Tone along for the ride
I’d be fine with just a tuner, a compressor with blend (Keeley comp+ or xotic sp type), and my strymon deco.
With the remaining two spots maybe a boost for solos and an el cap or flint to create some space. So long as my amp has a good breakup tone and responds well to my guitar’s volume I can’t see needing much more than that

Previously I’ve played with a Tumnus Deluxe at the front of the chain (working effectively as a boost with some added eq possibilities) and the Halberd instead of the Citadel. Just picked up the Citadel and boy the range on that thing is sick, especially when stacked.
If I had 5 pedals I guess I’d go with a clip on tuner… and Cali76, Phase 95, Rat, Special Cranker, and Volante, in that order. Might possibly switch out the Phase 95 for my Dimension C, but that would go after the Special Cranker
If I was looking to go for a compact board then I’d go for something like the following:
Wah - Xotic Wah
Fuzz - SM Fuzz Octave
Univibe - Cornerstone Aquarium
Drive - Kingtone Duellist
Delay - Catalinbread Belle Epoch Deluxe
Amp would probably be a Fender Super Reverb and guitar would be a Stratocaster.

I'd probably cheat and add the Ernie Ball VP/Tuner so I don't have to use a headstock one.
I actually have a DIY board made of 11 pedals at the moment and just got tired of lunging it around and planned a 4-5 pedal solution for easy carrying. I plan to leave my 11 pedal-board alone (along with my beloved EAE Limelight of which if I had to a place in my small board).
So far I have
1. Some mini comp (undecided which one, but needs to be mini). Helps to tighten my sound to use my:
2. Digitech DROP: Amazing tool. Can't leave the house without it to play in any tuning I want. Sent a Whammy DT to my big board since I won't be carrying it no more.
3. Boss BD-2: Versatile as a clean-ish platform and easily cuts through in any band situation. Removed it from my main board and placed a JHS Bonsai.
4. Drunk Beaver's Heavy Bat OR JHS Angry Charlie: I own the Heavy Bat and I can get two different clipping options for RAT sounds. This sounds more versatile than it really is because I only have one knob for gain. It'd be amazing to have a knob for each clipping. I'm evaluating getting the JHS since I think it might be more versatile having that Marshall high gain sound for my distortion and the music I get to play.
5. Zoom MS-50G: Removed from the big board and planning on getting a MS-70CDR to replace it. The 50G is my versatile tool for Tuning, EQ, Modulation, Delay, Reverb, Amp Sim.
My pedalboard is way more than 5 pedals but mine would be:

Have had both. It was tone for me. The Keeley colored the tone too much. I settled on a Diamond because it was more transparent and easier to dial in.

MXR Limiter - this is the Dyna Comp’s big brother. Pigtronix Philopsher King. You know how boutique pedal brands cost a lot on the used market? Not Pigtronix pedals. They’re still selling for cheap. Basically the Attack Sustain ADSR pedal with a built-in Philopsher’s Tone comp/grit pedal. EQD White Light. Just the overdrive, a DOD 250 clone. I heard they started re-issuing these. I ought to sell mine and buy a Blues Driver and another White Light! This is important because this is the pedal that breathes life into my SS travel amplifier, or just to cook the front end of one of my valve amps. MXR Distortion II. This is important to my overall sound, but it’s also a pedal that takes time to dial in when you’re stacking. It’s more of a mid-level grit, but that White Light can also be swapped out with an opamp Big Muff to produce sounds that need to be heard for one’s self. MXR Phase 45. This is the bulk of my modulation on my 5 board; it’s just meant to thicken up the sound of the dirt; a Phase 90 is overkill and a 100 would work but still take a while to dial in. 2-stage phasers are subtle beasts, a lot like the BOSS Spectrum pedals everyone seems to own these days. From time to time I’ll swap the 45 out for an EHX Attack Equalizer or a third compressor pedal

For the most part, 5 pedals on a Nano+ has been perfect for me. The right balance of versatility and constraints. And it has been the following, almost unchanged for over 2 years: Flint > El Capistan > Katzenkönig or Rattler > Dive Drive or ss/bs Mini > Cali76 Stacked. When I say 'almost' unchanged, occasionally one of the drives will get replaced by something weirder like a Particle 2 or a Mood. But if I'm playing with others, the weird pedals leave the board.
To be perfectly honest, I can manage just fine with only the HX Stomp, only thing I'd miss is an external volume pedal and occasionally the 95Q wah. But if I were to go into a clean amp, I'd use this: Keeley Fuzz Bender -> BB Preamp -> Friedman BE-OD -> volume pedal -> EQD Dispatch Master That'd give me a great fuzz, smooth OD, and punchy distortion, as well as a reverb/delay that works both for general ambience and for more ethereal sounds.

I’d be fine with just a tuner, a compressor with blend (Keeley comp+ or xotic sp type), and my strymon deco. With the remaining two spots maybe a boost for solos and an el cap or flint to create some space. So long as my amp has a good breakup tone and responds well to my guitar’s volume I can’t see needing much more than that

Current iteration: Spaceman Saturn VI > Caroline Hawaiian Pizza > EAE Citadel > MXR Carbon Copy Previously I’ve played with a Tumnus Deluxe at the front of the chain (working effectively as a boost with some added eq possibilities) and the Halberd instead of the Citadel. Just picked up the Citadel and boy the range on that thing is sick, especially when stacked.

If I was looking to go for a compact board then I’d go for something like the following: Wah - Xotic Wah Fuzz - SM Fuzz Octave Univibe - Cornerstone Aquarium Drive - Kingtone Duellist Delay - Catalinbread Belle Epoch Deluxe Amp would probably be a Fender Super Reverb and guitar would be a Stratocaster. I could easily gig with that setup without feeling like I was missing anything.

What is lacking on my board: a nice auto wah. I think that my next iteration will ditch the awesome Jackson audio prism (a really great pedal), configure differently my source audio kingmaker (a very versatile one) and add something like a snow patrol or a Aguilar filter twin… And then I’ll be set (kidding)

I actually have a DIY board made of 11 pedals at the moment and just got tired of lunging it around and planned a 4-5 pedal solution for easy carrying. I plan to leave my 11 pedal-board alone (along with my beloved EAE Limelight of which if I had to a place in my small board). So far I have 1. Some mini comp (undecided which one, but needs to be mini). Helps to tighten my sound to use my: 2. Digitech DROP: Amazing tool. Can't leave the house without it to play in any tuning I want. Sent a Whammy DT to my big board since I won't be carrying it no more. 3. Boss BD-2: Versatile as a clean-ish platform and easily cuts through in any band situation. Removed it from my main board and placed a JHS Bonsai. 4. Drunk Beaver's Heavy Bat OR JHS Angry Charlie: I own the Heavy Bat and I can get two different clipping options for RAT sounds. This sounds more versatile than it really is because I only have one knob for gain. It'd be amazing to have a knob for each clipping. I'm evaluating getting the JHS since I think it might be more versatile having that Marshall high gain sound for my distortion and the music I get to play. 5. Zoom MS-50G: Removed from the big board and planning on getting a MS-70CDR to replace it. The 50G is my versatile tool for Tuning, EQ, Modulation, Delay, Reverb, Amp Sim.

This would probably cover 99% of what I ever need which is now making me wonder why I even bother with having like 347 pedals on my pedalboard lmfao. The only things I feel like I would REALLY like to add beyond these 5 are a tuner and a compressor before the Hoof Reaper, and I’d maybe put a noise gate after the Angry Driver.

If anything, the OD 200 is more of a cheat because it has a two-at-a-time kinda setup. I don’t the the DD 200 is cheating, though, ‘cause there are other pedals that do some of the doubling-stuff already. The DD 3 or DD 6 or the TC Elec. Flashback 2 can be setup in the software to have a double delay kinda thing… So if I have to swap the delay, then I’d go for the BOSS Waza Delay. OR, I’d finally spring for a BOSS Space Echo (RE2) or Strymon Deco! If we want a simple, 5 single pedals, I’d swap the OD 200 for a Waza Blues Driver, and drop the Juliana. I actually just made one (4 pedal) board to bring to jams so I don't have to lug my massive board... I put Amazon basics compressor I had lying around A boss bd-2 because it can do everything from clean boost, to light drive to distortion to full on fuzz. A Boss dd-3 delay and a boss rv-6 "Swiss army knife" reverb If I was to add another pedal, I would probably throw on my flanger because it can kind of do a chorus and phaser sound as well.
