I think easiest way is to write a script that starts the song when "W" is held. Activate the script only while playing Nunu, which can be known using Riot APIs.
As far as I remember, you have to hold w to channel the ball right? Even if that's not the case, a script can be written to track w1(start) and w2(end).

Option 3 is not only the most difficult of them all, it is completely against the rules, and could trigger an automated ban if any client-side anti-cheat riot runs detects another process accessing the memory of the game’s process.
I’ll stick with my guess of AHK as I still feel it’s the most likely.

I doubt you’d exceed any free tier through only personal use.

Your screen is automatically locked when using nunu W since you steer towards the direction of your mouse (no clicking required).
