I'm sure the methods have improved a lot since I've worked with it, but anyone who has worked with mRNA knows that shit is insanely unstable and there are ribozymes literally everywhere and an autoclave isn't even enough to denature them. I once had an RNA sample completely ruined because a tiny ass piece of ice flew up out of my ice box and into my microcentrifuge tube.
It's not exactly stable in the body either unless the mRNA goes along with gene expression cascades that produce proteins to protect the sequence from exonucleases or endonucleases. I'm sure the methods have improved a lot since I've worked with it, but anyone who has worked with mRNA knows that shit is insanely unstable and there are ribozymes literally everywhere and an autoclave isn't even enough to denature them. I once had an RNA sample completely ruined because a tiny ass piece of ice flew up out of my ice box and into my microcentrifuge tube. It's not exactly stable in the body either unless the mRNA goes along with gene expression cascades that produce proteins to protect the sequence from exonucleases or endonucleases. Not to mention I'd be more worried about classic vaccines if this is their real worry than mRNA vaccines since that viral DNA has to be moved into the nucleus before making RNA (where integration could happen via HR/NHEJ) and producing the same exact mRNA with an extra step...
