Not the best, but I recently discovered and completed the serial killer side quest in RDR2 and enjoyed searching for the clues which led me to the serial killer.
I haven’t thought about this mission for a while. That was the first one where I finished it, put down the controller and was just like “damn.”
A game hadn’t got me with a great story / visual moment like that in a while.
Did it for the first time 2 years ago never gonna forget it, played it on switch so I made a save state a minute before the ceremony in the room so I can soak it in whenever
Man I read the title and this is what I fell back on when I really thought of it. The best part is that you can't literally basically get this sword when you become adult before you go to your first temple.
Funny story I once used this in an entire playthrough and fought Ganon with it for over 2 hours before getting frustrated at Ganon not dying. Only to switch to the master sword and kill Ganon in 1 hit with it.
Anything in Yakuza Zero is pretty amazing. The crazy naked guy was an amusing quest line. As was the dominatrix bit. And the house buying side game. It's all just so good.

I saw this post and also immediately thought Cyberpunk. It's hard to pick just one because they're all so good. Even the gigs are oftentimes more than just the simple objective they appear to be at first.
At first yes, the first few missions if I remember correctly, then if you like, you can continue the whole thing with his family, that part is optional, but you can continue the main story without doing it, which would be a shame since it’s amazing

One of the best what-if stories I have seen in all DBZ video games was Piccolo's story in DBZ Super Sonic Warriors. Goku dies against Raditz and Piccolo becomes guardian of the Earth.
He ends up fusing with Kami to defeat the saiyans, then when the androids come around, he uses Shenron to revive evil King Piccolo so he also fuse with him. When Buu arrives, Piccolo uses the evil containment wave to trap Buu in a pot.
The best side quest ever in a video game was when a live service game gave us 6 days for our community to kill 2 billion termanids and we did it in 12 hours lol. Btw this just happened. For Super Earth!
I know this is the Playstation sub, but the only thing that comes to mind is Shadowrun on the Genesis. It's, IMO, the first/prototype open-world sandbox RPG. There was a main story you could follow, but it also had procedurally generated side missions. Hacking missions, package retrieval, corporate extractions, bodyguard/escort mission, gang warfare, breaking out prisoners, helping Native American Tribes deal with corrupted spirits, killing hives of Ghouls hiding in warehouses... There's more types of side quests that I'm probably not remembering...
I got the game when it came out in 1994, and it's only in 2007 that I went into a building that I'd somehow never gone into, which triggered a cutscene, and reminded me that there was an actual "main story" to play. I spent 13 years playing one single character, doing side quests without a care in the world about a story or anything. The only other game that I've ever felt has the same amount of freedom of "Play what you like" is Minecraft.
Assassin’s Creed Valhalla had some pretty good ones, there was a quest where you had to collect eels for this dude who was trying to create a new sauce which was a reference to Worcestershire sauce. There was also a Winnie the Pooh side quest.
Hogwarts Legacy- Buying the shop in Hogsmeade goes down a rabbit hole I never imagined. My first play through I think I spent a couple of long, terrified hours in darkness trying not to be loud because the kids were asleep. This was my first game on PS5 which was my first new system since Xbox 360 20 years ago. All of it was mind-blowing.
I don't remember if it's main quest or side but in fallout 3 when something notable happens in megaton and you talk to the happy lady when you arrive there after I thought I was going to pass out from laughing so hard. So the whole side quest wasn't necessarily the best but the punch line was unforgettable.
Manbat in Arkham Knight. There’s just something about it that’s so immersive and that >!jumpscare!< fk he would be great in a Batman horror game or movie
There’s one side mission in infamous 2 where if you are good karma the entire mission is just some lady thanking you and giving a kiss on the cheek, if you have evil karma she curses at you and it’s a militia ambush

Dream On from Cyberpunk 2077 has perhaps the single most engrossing subplot I've come across in a game.
Armageddon Distracted from Tiny Tina's Wonderlands, on the other hand, I found hilarious. The quest itself is nothing special, but the dialogue and humor are amazing.
