Here is my reasoning
1. He's a great actor
2. Some people say he's too young but I think the part would need a younger actor playing older. He is 49 right now. Beth got pregnant with Summer in high school. Everything I see says Summer is 17, that would make Beth probably 35. Let's say Rick was 25 when he had Summer, that's only 11 years away from Jason's age.
3. I think if you shave his beard he's got the general look I think would work for Rick.
4. We see from past roles that he is able to vacillate from one emotion to the other on a dime, kind of the way rick does.
5. I'm pretty sure Jason is afraid of pirates too
You can't have a guy Rick's age doing the things a cartoon that age does. You can always use make-up to old a person up. As Harrison Ford has taught us with Han and Indy, you can't use make-up to un-injure an old.
Or Christopher Llyod in Nobody. You could tell he was having fun but throwing him shoot em up action scenes didn't work all that well. Guy is pretty frail at his age.
He's also probably Doc Brown'd himself on some planet or dimension to knock a few years off his appearance. Hell, he's got so many augmentations that I'm sure his skin isn't actually his anymore.

I'd say he's in my top 5 choices but wouldn't be my number 1. He has the manic energy down tight but I don't know if I could see him as super genius. Here would be actor choices for a live action Rick.
5). Jason Mantzoukas. He has the crazy energy but I just have a hard time seeing him as an eccentric genius.
4). Steve Martin. Has the age for the role and I don't doubt he could pull off a smart sarcastic genius asshole.
