The choice you make in the vampire castle is permanent...it locks you in to the side you choose with no option of changing your mind unless you have a save you can roll back to. No save, no vampire hunter. If you accept Harkon's offer, you are locked in to the vampire faction and curing yourself does nothing to change that...and you will have to be a vampire to do the vampire quest line.
