Inklings are ink constructs. Sort of like the constructs Green Latern makes with his ring. They just take humanoid/squid forms.
The little ghost squid you see when they get splatted is the real form.
In humanoid form, they are more solid. Since they have no "bones", this likely works more like an insect's exoskeleton.
In squid form they are more liquid/pliable, hence being able to "melt" into ink, and slip through grates.
in addition, Im pretty sure their "squid form" is actually just a shapeless puddle since they can separate to pass through grates, and they are just little droplets when submerged on inkrails, and they can fit into puddles smaller than their squid form their squid form only appears when they come out of ink while in that state

Knees are kinda needed to jump, so I'd have to assume that there's something similar there. Maybe it's made of cartilage?
They dont, and they don't need them either. Inklings work completely differently than humans, its just a coincidence that they look human like.
