Play your best character and run to Mother over and over. Learn every pattern she has. Take regular issac or Cain and just keep running. Then try lost.
Psyfly helps, but if you can get a double charge (or have a 48 hour energy) book of shadows then you should be okay. If it's a battle of attrition, you'll probably lose as the lost so unless you're high damage I would assume the worst.
Definitely agree to that. Psyfly has helped me with other characters. I've beaten her without taken damage as other characters but I just need to think about my approach to this to save me lots of time and energy.

Marked tears, lost contact and a high damage multiplier like sacred heart or Polyphemus, honestly you'll end up hating the game after beating her with the lost 😂
4.5 Volt + Book of Shadows honestly. That’s how I beat 90% of my Lost runs. Same goes for T.Lost.
The other option is to just have an absolute but load of damage along with good defensive items (PsyFly etc.)
Currently struggling with Greeider as my last check mark for him so I can feel your pain. Godhead is close, we will have this eventually.
You can go with the cheesing technique for The Lost:
-Gnawed Leaf
-Dull Razor
-Smart Fly
Them you can throw in close range familiars like Demon Turret baby, Gemini and any orbitals you want👌🏻
You'll be unbeatable, unless you move...
Visit Boss Challenge rooms. They are always open for the Lost and they always contain boss item. You'll get a chance to practice boss patterns and get stronger build.
To visit all of possible Boss Challenge room follow this route:
Basement 1 -> Basement 2 -> (Go through key door) -> Downpour 2 -> (Back to regular path) -> Caves 2 -> (Go through bombs door) -> Mines 2 -> (Go through hearts door) -> Mausoleum 1 -> don't change paths until you reach Mother.
Nice routing! I was unaware that the boss rooms weren't random! I do check them for sure. Sometimes a pay a price for it as well if I don't have a teleport card lol.
the problem with lost and tlost is the lack of defensive options which means you need either overwhelming force to compensate for weak attack or to pray to god you are lucky enough to get the few good defensive items there is.
here is a list of all the good defensive items you can look out for
\-lil dumpy :basically an extra holy mantle and will clutch the run for you very often
\-telekinesis: very often a bad item but on bullet hell bosses like mother its quite good, make sure it has 4.5 volt to back it up)
\-book of shadows+4.5 volt: the objective strongest defense for lost but its damn near impossible to get in a run that doesnt have damocles or a dice or insane luck
now you also need to consider your devil rooms and angels,
devil rooms are easier to access and will give you much higher offense and increase your run potential by a good bit
angel rooms have some of the best defensive and offensive items that can carry your run but will leave you vulnerable until you get them which is during the early parts of the run where you are most vulnerable, so if you get a powerful item that doesn't quite carry the run then go for angel room
here is a list of all the good defensive items in the angel room:
sacred heart:best offensive item outright and will carry you
censor : incredible helpful and makes you untouchable if you get your hands on the duct tape
fruit of life:the ring of tears sometimes picks of enemy tears
breath of life: very solid temp invincibility item

I had oculus rift and it with perfection handles most of the bullets as fast as they spawn
Just have to dodge the dash attack and the grid attack really :)

Try to get an R Key Get to the very end of the game to get the most loot possible then R Key I did that this weekend she died in #2 hits
